• Dithiothréitol (TNT), or Cleland’s reagent, is a small molecule organic reducing agent.
• In its reduced state, DTT exists as a linear molecule, but upon oxidation, it forms a six-membered ring structure containing disulfide bonds.
• DTT is a potent reducing agent, largely due to the stability of its oxidized state, the six-membered ring with disulfide bonds.
• One application of DTT is as a reducing agent and deprotectant for thiol-modified DNA.
• Thiol-modified DNA tends to dimerize in solution, surtout en présence d'oxygène, which reduces the efficiency of certain coupling reactions, such as DNA immobilization in biosensors.
• The addition of DTT to DNA solutions can reduce dimerization, améliorer l'efficacité expérimentale.
• DTT is also commonly used to reduce disulfide bonds in proteins, preventing the formation of intra- or intermolecular disulfide bonds between cysteine residues.
• Cependant, DTT may not effectively reduce disulfide bonds buried within the protein structure (inaccessible solvent), which often requires denaturation of the protein.
• The rate of disulfide bond reduction in the presence of DTT can indicate the depth of burial of these bonds, allowing insights into protein folding.
