• Dithiothreitol (DTT), or Cleland’s reagent, ist ein kleines Molekül organischer Reduktionsmittel.
• In its reduced state, DTT exists as a linear molecule, but upon oxidation, it forms a six-membered ring structure containing disulfide bonds.
• DTT is a potent reducing agent, largely due to the stability of its oxidized state, the six-membered ring with disulfide bonds.
• One application of DTT is as a reducing agent and deprotectant for thiol-modified DNA.
• Thiol-modified DNA tends to dimerize in solution, insbesondere in Gegenwart von Sauerstoff, which reduces the efficiency of certain coupling reactions, such as DNA immobilization in biosensors.
