Product Overview
- Dithiothreitol (DTT), or Cleland’s reagent, is a small molecule organic reducing agent.
- In its reduced state, DTT exists as a linear molecule, but upon oxidation, it forms a six-membered ring structure containing disulfide bonds.
- DTT is a potent reducing agent, largely due to the stability of its oxidized state, the six-membered ring with disulfide bonds.
- One application of DTT is as a reducing agent and deprotectant for thiol-modified DNA.
- Thiol-modified DNA tends to dimerize in solution, especially in the presence of oxygen, which reduces the efficiency of certain coupling reactions, such as DNA immobilization in biosensors.
- The addition of DTT to DNA solutions can reduce dimerization, enhancing experimental efficiency.
- DTT is also commonly used to reduce disulfide bonds in proteins, preventing the formation of intra- or intermolecular disulfide bonds between cysteine residues.
- However, DTT may not effectively reduce disulfide bonds buried within the protein structure (inaccessible solvent), which often requires denaturation of the protein.
- The rate of disulfide bond reduction in the presence of DTT can indicate the depth of burial of these bonds, allowing insights into protein folding.
- Other names: 1,4-Dithio-DL-threitol, Threo-1,4-dimercapto-2, 3-butandiol
