Raccolta campione:
- Raccogliere 0.1-1 ml blood sample.
Preparazione del campione:
- For blood samples, aggiungere 2-3 times the volume of 1x erythrocyte lysate.
- Fully invert and mix well.
- Centrifugare a 12,000 giri al minuto per 1 minuto.
- Carefully remove the supernatant. The precipitate should be white or light red.
- Aggiungere 400 μl reagent buffer I and 10 μl proteinase K (10 mg/ml) to the precipitate.
- Vortex and mix for 15 secondi, then let it sit at room temperature for 2 minuti.
Special Case: Low-level Organisms:
- If processing blood from poultry, birds, or other low-level organisms where red blood cells contain nuclei:
- Skip adding 1x red blood cell lysate.
- Directly add 200 μl of reagent buffer I (total volume not exceeding 500 ml).
- Let it sit at room temperature for 2 minuti.
Digestion:
- Aggiungere 10 μl proteinase K (10 mg/ml) to the mixture.
- Thoroughly mix by inversion.
- Digest at 65°C for 10 minuti.
- Invert and mix 6-7 times during digestion until complete.
Transfer to Reagent Plate:
- Add the lysate from the previous step to the 96-well reagent plate.
- Follow specific steps outlined in the automatic extraction protocol for further processing.
