Protocole (pour référence seulement)	Tissue Preparation: Clean fresh tissue with absolute PBS multiple times. Fix immediately (See Note 1). Dehydration with Sucrose-PBS Solution, 30%: Submerge tissue in 10-20 times its volume of Sucrose-PBS Solution, 30%. Dehydrate at 4°C. Replace sucrose solution every 12-24 heures. Once tissue completely sinks, remove it. Embedding and Further Operations: Proceed with subsequent embedding and other operations as per experimental requirements. Note: Fixation of tissues of different sizes: Small samples (less than 5mm thick) or isolated embryos: Fix with G2161-Neutral Buffered Formalin, 10%, or P1110-Paraformaldehyde Fixative, 4%, pour 1 heure. Wash with PBS. Large samples (5-20mm thick): Recommended to fix at 4°C for at least 8 heures. Wash with PBS. Extra-large samples (more than 20mm thick): Perfusion fixative recommended. After in-situ fixation, remove samples and wash with PBS. Précautions: Sucrose solution prone to contamination. Maintain aseptic operations to prevent microbial contamination. Facultatif: Combination with G2461-Sucrose-PBS Solution, 20%, and G2462-Sucrose-PBS Solution, 10%, for complete dehydration while preserving tissue structure. Dehydration Steps: Immerse in sucrose PBS solution sequentially from low to high concentration until completely sinking to the bottom. Safety Measures: Wear experimental clothing and disposable gloves for safety and health.
