  • Tissue Preparation:
    • Clean fresh tissue with absolute PBS multiple times.
    • Fix immediately (See Note 1).
  • Dehydration with Sucrose-PBS Solution, 30%:
    • Submerge tissue in 10-20 times its volume of Sucrose-PBS Solution, 30%.
    • Dehydrate at 4°C.
    • Replace sucrose solution every 12-24 ชั่วโมง.
    • Once tissue completely sinks, remove it.
  • Embedding and Further Operations:
    • Proceed with subsequent embedding and other operations as per experimental requirements.

    • Fixation of tissues of different sizes:
      • Small samples (less than 5mm thick) or isolated embryos:
        • Fix with G2161-Neutral Buffered Formalin, 10%, or P1110-Paraformaldehyde Fixative, 4%, สำหรับ 1 ชั่วโมง.

      • Large samples (5-20mm thick):
        • Recommended to fix at 4°C for at least 8 ชั่วโมง.

    • Immerse in sucrose PBS solution sequentially from low to high concentration until completely sinking to the bottom.

    • Wear experimental clothing and disposable gloves for safety and health.
