  • Phosphate-Buffered Saline (PBS):
    • Commonly known as PBS
    • Frequently used phosphate buffer solution
    • Consists of:

      • Potassium dihydrogen phosphate
    • Lacks calcium and magnesium
    • pH value: 7.4
    • Commonly employed for:
      • Cell washing during cell culturing or other cell-related procedures
  • Dulbecco’s Phosphate Buffer Solution (D-PBS):
    • Variation of PBS lacking calcium and magnesium

  • Sucrose-PBS Solution, 30%:
    • Composizione:
      • Sucrose and PBS
    • Usage:
      • Precipitates fresh tissue to remove excess water
      • Prevents formation of ice crystals upon freezing
    • Sucrose serves as a dehydrating agent
Protocollo (solo per riferimento)
  • Tissue Preparation:
    • Clean fresh tissue with absolute PBS multiple times.
    • Fix immediately (See Note 1).
  • Dehydration with Sucrose-PBS Solution, 30%:
    • Submerge tissue in 10-20 times its volume of Sucrose-PBS Solution, 30%.
    • Dehydrate at 4°C.
    • Replace sucrose solution every 12-24 ore.
    • Once tissue completely sinks, remove it.
  • Embedding and Further Operations:
    • Proceed with subsequent embedding and other operations as per experimental requirements.
  • Nota:
    • Fixation of tissues of different sizes:
      • Small samples (less than 5mm thick) or isolated embryos:
        • Fix with G2161-Neutral Buffered Formalin, 10%, or P1110-Paraformaldehyde Fixative, 4%, per 1 ora.
        • Wash with PBS.
      • Large samples (5-20mm thick):
        • Recommended to fix at 4°C for at least 8 ore.
        • Wash with PBS.
      • Extra-large samples (more than 20mm thick):
        • Perfusion fixative recommended.
        • After in-situ fixation, remove samples and wash with PBS.
  • Precauzioni:
    • Sucrose solution prone to contamination.
    • Maintain aseptic operations to prevent microbial contamination.
  • Opzionale:
    • Combination with G2461-Sucrose-PBS Solution, 20%, and G2462-Sucrose-PBS Solution, 10%, for complete dehydration while preserving tissue structure.
  • Dehydration Steps:
    • Immerse in sucrose PBS solution sequentially from low to high concentration until completely sinking to the bottom.
  • Safety Measures:
    • Wear experimental clothing and disposable gloves for safety and health.
