Pre-processing of bacterial sample genomic DNA extraction 96 Uzorci

Instruction：

1. Sample Collection:

   • Collect 0.1-1 ml blood sample.

2. Priprema uzorka:

   • Dodati 2-3 times the volume of 1x erythrocyte lysate to the blood sample.
   • Fully invert and mix well.
   • Centrifuga na 12,000 RPM za 1 minuta.
   • Carefully remove the supernatant. The precipitate should be white or light red.
   • Dodati 400 μl reagent buffer I and 10 μl proteinase K (10 mg/ml) to the precipitate.
   • Vortex and mix for 15 sekundi, then let it sit at room temperature for 2 minuta.

3. Special Case: Low-level Organisms:

   • If processing blood from poultry, birds, or other low-level organisms where red blood cells contain nuclei:
   • No need to add 1x erythrocyte lysate.
   • Directly add 200 μl of reagent buffer I (total volume not exceeding 500 μl).
   • Let it sit at room temperature for 2 minuta.

4. Digestion:

   • Dodati 10 μl proteinase K (10 mg/ml) na smjesu.
   • Thoroughly mix by inversion.
   • Digest at 65°C for 10 minuta.
   • Invert and mix 6-7 times during digestion until complete.

5. Transfer to Reagent Plate:

   • Add the lysate from the previous step to the 96-well reagent plate.
   • Follow specific steps outlined in the automatic extraction protocol for further processing.
