Ekstrakcijski postupak:

1. Deparaffinization: Xylene removes paraffin wax, a common component used to preserve tissues.
2. Lysis and Digestion: A special solution (lysate) and an enzyme (protease) break down cells and proteins, releasing nucleic acids (DNA and RNA) into the solution.
3. De-Crosslinking: Heating at 90°C reverses the effects of formalin fixation, allowing easier access to nucleic acids.
4. Binding to Silica Membrane: After adjusting conditions with ethanol, the sample is transferred to a column. DNA/RNA binds to the silica membrane, while proteins pass through.
5. Washing: Buffers remove impurities like proteins and salts from the captured nucleic acids.
6. Elution: A low-salt buffer elutes the purified DNA/RNA, making it ready for downstream applications.

Skladištenje i stabilnost

• Proteinaza K: Keep it cool in the refrigerator (2-8°C) upon arrival for maximum effectiveness.
• Short-Term Storage (do 12 tjedni): Proteinase K can last at room temperature (15-25°C) for up to 12 weeks without issues.
• Everything Else: Store other kit components at room temperature (15-25°C). They are stable for at least 18 mjeseci pod ovim uvjetima.
• Important: If storing the entire kit cold, remember to warm up the buffers before use. All buffers should be at room temperature for optimal results.
