原則

• HiPure Plasmid Procedure:
  • Based on alkaline lysis of bacterial cells followed by adsorption of DNA onto silica in the presence of high salt.
  • Utilizes a unique silica membrane that replaces glass or silica slurries for plasmid DNA minipreps.
• Procedure Steps:
  1. Preparation and Clearing of Bacterial Lysate:
     • The alkaline method is used to prepare and clear bacterial lysate.
  2. DNA Binding:
     • Supernatant transferred to a column to bind DNA to the silica membrane.
  3. Washing and Elution:
     • Proteins and other impurities washed away.
